(TIF) (11.5 MB) Whole mount X-gal staining patterns of various organs of newborn mice reveal strong and homogeneous Cre activities in the lung. Lung (a-c), brain (d-f), mesenteric vessels (g-i), kidney (j-l), blood vessels in the thoracic wall (m-o), and liver (p-r) from control (a, d, g, j, m and p), L1cre(+);R26R (b, e, h, k, n and q), and B1cre(+);R26R (c, f, i, l, o and r) newborn pups were stained with the X-gal staining solution overnight. Blue regions represent the cells at which the cre recombinase is (was) expressed, except for the small intestine (g-i) and kidney (j-l) where endogenous â-galactosidase activities were present. X-gal staining pattern of the lung in L1cre;R26R (b) is the indistinguishable from that in B1cre mice (c). Brain is another organ where L1cre activity was readily detected at the same level as B1cre activity (e, f). X-gal staining patterns in mesenteric vessels, kidneys and blood vessels in the thoracic wall (as a representative of blood vessels in skeletal muscles) show that L1cre has patchy, mosaic cre activities in systemic vessels. Histological sections confirmed that most of X-gal positive cells are vascular endothelial cells (not shown). In the liver of L1cre mice, almost no X-gal-positive cells were detected.
