(TIF) (9.51 MB). EPCs versus mature endothelial cells. (a) We performed dual CD34 and KDR FACS on OECs and GEAECs with 4 different cells of different donors. Representative figures showing greater CD34+ and KDR+ double positive cells in OECs than in mature endothelial cells in the basal condition as well as the VEGF-stimulated condition. (b) ICAM-1 and αvβ5 were also more highly expressed on OECs than on GEAECs. (n=4) (c) Telomeric repeat amplification protocol (TRAP) was done with early EPCs, OECs, and GEAECs. 293 cells were used as control cells. (d) Telomerase activity, demonstrated as the total product generated (TPG), was significantly higher in the OECs than in the GEAECs. (e) In vivo, we transplanted OECs and GEAECs into ischemic muscle and a Matrigel plug. OECs, compared with GEAECs, showed greater proliferation and therefore resulted in more capillaries structures by day 21. When we performed immunofluorescent staining of DiI (red)-labeled these 2 kinds of cells with anti-KDR antibody (green), we could see that a greater number of OECs were stained than GEAECs, and also in a diffuse pattern in contrast to the speckled pattern of GEAECs.
