Supplemental Figures (Microsoft PowerPoint)
A: Double staining for β-gal and α-smooth muscle actin in human advanced atheroma. Granular blue stainings were co-localized with an immunoreactivity for α-smooth muscle actin (blown). Original magnification: ×1000.
B: Cell growth after infection with TRF2 D/N. Human endothelial cell populations infected with TRF2 D/N or Mock were purified with 1 μg/mL puromycin for 3 days and 25×104 cells were plated at Day 0. Cell number was then counted at indicated time points (n=4).
C: Western blot analysis for CDK inhibitors. The expression levels of p21 and p16 were significantly increased in TRF2 D/N-infected cell populations at Day 4.
D: Senescence-associated β-gal activity in HAEC infected with TRF2 D/N and Mock. Increased β-gal activity was observed in TRF2 D/N-infected cell populations (middle and right) as compared with mock-infected populations (left). Note that the blue staining in senescent cells in vitro resembles that in vivo (right).
