Shown in the figure are schematic diagrams of voltage pulses imposed on HEK 293 cells expressing I1768V channels (upper traces) and the resulting whole cell currents recorded in the absence (control, A) and presence (TTX, B) of TTX, as well as after returning to TTX-free conditions (washout, D). In the absence of TTX, currents in response to the negative voltage ramp consist of a transient inward current (I1768V channels reopening, arrow) superimposed on a current that is linearly proportional to membrane potential. This linear background current which is TTX-insensitive is seen in panel B. Digital subtraction of the TTX-resistant background current in I1768V channel Iramp (arrows) in control conditions (panel C) and after washing out TTX (panel E).
